ABSTRACT

Abstract Experimental models that consider host-pathogen interactions are relevant for improving knowledge about oral candidiasis. The aim of this study was to assess the epithelial immune responses, Candida penetration of cell monolayers, and virulence during mixed species culture infections. Single species cultures of Candida albicans and mixed cultures (C. albicans, Streptococcus mutans, and Streptococcus sanguinis) were used to infect monolayers of HaCaT and FaDu ATCC HTB-43 cells for 12 h. After infection, IL-18 and IL-34 gene expression was measured to assess epithelial cell immune responses, and lactate dehydrogenase (LDH) activity was measured as an indicator of cell damage. Microscopy determined C. albicans morphology and penetration of fungal cells through the keratinocyte monolayer. Monolayers devoid of infection served as controls. Data were analyzed by an ANOVA one-way test followed by Tukey's post-hoc test (α = 0.05). The results found that IL-18 and IL-34 gene expression and LDH activity were significantly (p < 0.05) upregulated for both cell lines exposed to mixed species cultures compared with C. albicans alone. Candida albicans yeast and hyphae were evident in C. albicans only infections. In contrast, monolayers infected by C. albicans, S. mutans, and S. sanguinis exhibited higher microbial invasion with several hyphal aggregates detected. The presence of streptococci in C. albicans infection enhances the virulence and pathogenicity of the fungus with associated increased immune responses and tissue damage. Extrapolation of these findings to oral infection would indicate the added potential benefit of managing bacterial components of biofilms during treatment.


Resumo O objetivo deste estudo foi avaliar a resposta epithelial imune, a colonização da Candida albicans em monocamadas celulares e sua virulência em resposta a infecções de culturas de biofilme multiespécie. Culturas de biofilme monoespécie de C. albicans e culturas mistas (C. albicans, Streptococcus mutans e Streptococcus sanguinis) foram utilizadas para infectar monocamadas de células HaCaT e FaDu por 12 h. Após a infecção, a expressão dos genes IL-18 e IL-34 foi medida para avaliar as respostas imunes das células epiteliais. A atividade da lactato desidrogenase (LDH) foi medida como um indicador de dano celular. A microscopia determinou a morfologia de C. albicans e a penetração das células fúngicas através da monocamada de queratinócitos. Monocamadas em que não houve infecção serviram como controles. Os dados foram analisados por um teste ANOVA one-way seguido pelo teste post-hoc de Tukey (α = 0,05). Os resultados demonstraram que a expressão gênica de IL-18 e IL-34 e a atividade de LDH foram (p < 0,05) reguladas positivamente para ambas as linhagens de células expostas a culturas de espécies mistas em comparação com C. albicans isoladamente. Leveduras de C.albicans e hifas foram evidentes em infecções apenas por C. albicans. Entretanto, monocamadas infectadas por C. albicans, S. mutans e S. sanguinis exibiram maior invasão microbiana com vários agregados de hifas detectados. Dessa maneira, a presença de estreptococos na infecção por C. albicans aumentou a virulência e a patogenicidade do fungo com respostas imunes aumentadas associadas a danos nos tecidos. A extrapolação desses achados para a infecção oral indicaria o potencial benéfico do controle dos componentes bacterianos em biofilmes durante a terapia da candidíase

ABSTRACT

Peste des petits ruminants (PPR) is a notifiable and reportable disease recognised by the Office Internationale Epizooties (OIE) and penned for eradication by 2030. It is an endemic and devastating disease of small ruminants in some countries of Africa, Asia, and the Middle East. The disease militates against small ruminant production and increased flock numbers in the region. Information and literature on the current review was especially obtained and compiled from CABI abstract data base, PubMed, Scopus, Elibrary USA and other online publications. This review highlights the viral structure, epidemiology, predisposing factors, clinical signs, pathology, immunoprophylaxis, diagnostic techniques and therapeutics of the disease with a view to generating increased scientific research interests on PPR. It is hoped that more robust scientific research breakthroughs could emerge particularly in the area of immunoprophylaxis, rapid penside diagnosis, and alternative therapeutics such as the use of probiotics. This will in no small measure boost small ruminant production, alleviate poverty and increase food security in countries endemic with the disease.

Resumen Las cepas de Escherichia coli productoras de toxina Shiga (STEC) son reconocidas como responsables de un alto número de casos de enfermedades de transmisión alimentaria a nivel mundial. Su patogenicidad ha sido vinculada directamente con la actividad de las toxinas (Stx); sin embargo, la habilidad de estas bacterias para colonizar al huésped y otras superficies puede ser esencial para desarrollar su poder patogénico. La gran plasticidad genómica de cepas STEC se infiere de la variabilidad de perfiles de virulencia, con la frecuente emergencia de cepas con nuevos genes, codificados en nuevas islas de patogenicidad vinculadas al metabolismo y la adherencia. La formación de biofilm es un mecanismo espontáneo por el cual las cepas STEC resisten en un ambiente hostil, lo que les permite sobrevivir y, de esa forma, llegar al huésped, a través de los alimentos o de las superficies que están en contacto con ellos. Este mecanismo presenta una alta variabilidad intra e interserotipo y su desarrollo no depende solo de los microorganismos que lo conforman. Factores inherentes al ambiente (pH, temperatura) y la superficie (acero inoxidable, poliestireno) a la que pueden adherirse influyen en la expresión de biofilm. El concepto «una salud¼ implica la interrelación entre los actores de salud pública, animal y ambiental para lograr alimentos inocuos y evitar contaminación cruzada y resistencia a sanitizantes, lo cual pone de manifiesto la necesidad de identificar patógenos emergentes a través de nuevos marcadores moleculares, que detecten cepas STEC portadoras del denominado locus for enterocyte effacement (LEE) o del locus de adherencia y autoagregación (LAA).


Abstract Shiga Toxin-producing Escherichia coli (STEC) is recognized as being responsible for a large number of foodborne illnesses around the world. The pathogenicity of STEC has been related to Stx toxins. However, the ability of STEC to colonize the host and other surfaces can be essential for developing its pathogenicity. Different virulence profiles detected in STEC could cause the emergence of strains carrying new genes codified in new pathogenicity islands linked to metabolism and adherence. Biofilm formation is a spontaneous mechanism whereby STEC strains resist in a hostile environment being able to survive and consequently infect the host through contaminated food and food contact surfaces. Biofilm formation shows intra-and inter-serotype variability, and its formation does not depend only on the microorganisms involved. Other factors related to the environment (such as pH, temperature) and the surface (stainless steel and polystyrene) influence biofilm expression. The «One Health¼ concept implies the interrelation between public, animal, and environmental health actors to ensure food safety, prevent cross-contamination and resistance to sanitizers, highlighting the need to identify emerging pathogens through new molecular markers of rapid detection that involve STEC strains carrying the Locus of Enterocyte Effacement or Locus of Adhesion and Autoaggregation.

Background: Much controversies have been associated with the pathogenicity of Mycoplasma hominis but little has been done to unravel the mystery behind the different views. This study aimed at investigating the genetic variants abounding within M. hominis and the distribution of the virulent genes among the variants. Methodology: Twenty (20) M. hominis isolates from high vaginal swabs of women (11 from pregnant women and 9 from women presenting with infertility) attending the Obstetrics and Gynaecology clinics of Nnamdi Azikiwe University Teaching Hospital (NAUTH), Nnewi, Nigeria, were sequenced using 16S rRNA universal gene target for the purpose of phylogenetic analysis and epidemiological typing. The isolates were also screened for the presence of M. hominis variable adherence antigen (vaa) and p120 virulent genes using primer constructs from the respective genes in a conventional PCR protocol. Results: Of the 20 M. hominis vaginal isolates, 4 phylogenetic strains were detected; strain MHS43 constituted 10/20 (50.0%) [2/9 (22.2%) from infertile women and 8/11 (72.7%) from pregnant women]; strain MHBS constituted 3/20 (15%) [3/9 (33.3%) from infertile women and 0/11 (0%) from pregnant women]; strain MHSWP2 constituted 4/20 (20.0%) [3/9 (33.3%) from infertile women and 1/11 (9.1%) from pregnant women]; while strain MHKC87 constituted 3/20 (15%) [1/9 (11.1%) from infertile women and 2/11 (18.2%) from pregnant women].Each of vaa and p120 genes was detected in 14 of 20 isolates, while 6 isolates did not carry the genes. A 2-way ANOVA test showed that none of the genes was significantly associated with a particular strain (p=0.8641). Conclusions: The different views regarding the pathogenicity of M. hominis may be linked to the heterogeneity within the species and lack of homogeneity in the virulent genes as witnessed both in the intra species and intra strain levels.

@#Abstract: Objective To investigate the prevalence and pathogenic characteristics of Yersinia enterocolitica infection in children with diarrhea under 5 years of age in western Yunnan, and to provide a basis for the prevention and treatment of infectious diarrhea in children. Methods Feces were collected from under five-year-old children with diarrhea in the First Affiliated Hospital of Dali University from 2020 to 2021. Clinical information of the cases was also collected. Yersinia enterocolitica was isolated from the samples after cold enrichment on selective culture plates, and the pathogenic characteristics of Yersinia enterocolitica were analyzed by biological type and serotype and virulence gene detection. Results A total of 397 feces were collected. Seven strains of Yersinia enterocolitica were isolated in three samples, and the prevalence of Yersinia enterocolitica infection was 0.76% (3/397). Among the three positive samples, two Yersinia frederiksenii or Yersinia intermedia were isolated in specimen No. 212 , and five Yersinia enterocolitica were detected in specimens No. 24 and 226. Two Yersinia enterocolitica isolated from one sample were biological type 1A, and the virulence gene test results were ail-/ystA-/ ystB+ /yadA-/virF-, which were non-pathogenic Yersinia enterocolitica. Three Yersinia enterocolitica isolated from the other sample were biological type 3, serotype O∶3 (rfbc+), and virulence gene detection results were ail+/ystA+/ystB-/yadA+ /virF+, which were pathogenic Yersinia enterocolitica. While pathogenic Yersinia enterocolitica was detected from feces of children with diarrhea at 11 months of age with a infection rate of 0.50%(2/397). Conclusion Sporadic infection of pathogenic Yersinia enterocolitica was found in under five-year-old children in western Yunnan Province. It is necessary to strengthen the monitoring and research of Yersinia enterocolitica.

@#Staphylococcus aureusis a major cause of hospital and community acquired infection,which can express many kinds of cell wall-related and extracellular virulence factors,causing a variety of infectious diseases. Ebh protein is involved in the adhesion,aggregation and biofilm formation of Staphylococcus aureus,and thus closely related to its strong pathogenicity. The pathogenesis of Staphylococcus aureus is controlled collaboratively by different two-component signal transduction systems and transcriptional regulation factors,among which ArlRS is the key two-component system(TCS)necessary for the adhesion,biofilm formation and virulence. At the same time,ArlRS regulates Ebh protein through global regulation factor MgrA,thus affecting the role of Staphylococcus aureus in human infectious diseases. In this paper,the regulatory mechanism was reviewed.

ABSTRACT

Objective:To study the molecular phylogeny and virulence gene profile of Francisella salimarina. Methods:Phylogenetic analysis of Francisella salimarina was performed based on the global genome data of related Francisella species on GenBank database. The consistency in phylogenetic analysis based on single marker genes (such as 16S rRNA gene, rpoB gene and mdh gene) and the core genome as compared. Virulence genes and antibiotic resistance genes were annotated using the virulence factor database (VFDB) and the Comprehensive Antibiotic Resistance Database (CARD), respectively. The virulence of Francisella salimarina was analyzed with a Galleria mellonella (greater wax moth) infection model using Francisella philomiragia ATCC 25015 T as reference strain. Results:The phylogenetic analysis revealed that Francisella salimarina was closely related to Francisella philomiragia. The phylogenetic tree based on mdh gene was highly similar to that based on the core genome. Francisella salimarina could be differentiated from other related species by 16S rRNA gene or mdh gene, with the latter being more accurate. Eight Francisella salimarina strains carried multiple virulence genes, mainly involved in secretion, adhesion, immune regulation, motility and stress survival. Moreover, beta-lactam resistance gene blaFPH was identified in all eight strains. Francisella salimarina showed high lethality in the Galleria mellonella infection model, which was similar to Francisella philomiragia ATCC 25015 T. Conclusions:Francisella salimarina was a rare pathogen with similar pathogenicity to Francisella philomiragia. The mdh gene could be used as a molecular target for rapid identification of Francisella salimarina.

ABSTRACT

Epilepsy is a common disease in nervous system, of which patients often present with spontaneous unpredictable spontaneous seizures. Sudden unexpected death in epilepsy (SUDEP) is one of the most serious complications of epilepsy, and it is also the main cause of premature death of epileptic patients. Generalized tonic-clonic seizures, age and genetic factors are common risk factors of SUDEP. This article summarizes the classification of SUDEP and epidemiology, mechanism, risk factors, risk assessment and preventive methods of SUDEP to help physicians to understand the difference between SUDEP and sudden cardiac death.

ABSTRACT

Bath towels are an important part of our everyday life, but they often amass a large number of micro-organisms which may sometimes be harmful to us. For the study a total of 10 samples plus a control were collected from bath towels of female students in NDDC hostel, post-graduate hostel and control from Mile 3 market respectively between November 2021 to February 2022. Standard methods were employed for the sampling and determination of microbiological characteristics. Identification of bacteria was carried out using colonial, morphological and biochemical characteristics. Statistical analysis were performed using the T-test method and sensitivity was carried out on the isolates to detect pathogenicity. Range of microbial counts of NDDC hostel were: Total heterotrophic bacteria 2.0 x 104 cfu/ml to 3.17 x 104 cfu/ml, Total coliform count 0.48 x 104 cfu/ml to 2.99 x 104 cfu/ml in the first sampling. Post graduate hostel ranged from: Total heterotrophic bacteria 1.48 x 104 cfu/ml to 0.18 x 104 cfu/ml in the first sampling. For the second sampling, NDDC hostel microbial count ranged from 0.73 x 104 cfu/l to 1.15 x 104 cfu/ml in the total heterotrophic bacteria, and 0.31 x 104 cfu/ml to 0.13 x 104 cfu/ml for total coliform count. Post graduate hostel ranged from 0.27 x 104 cfu/ml to 0.93 x 104 cfu/ml for total heterotrophic bacteria and 0.22 x 104 cfu/ml to 0.18 x 104 cfu/ml for total coliform count. The control microbial count for total heterotrophic bacteria were 0.12 x 104 and 0.16 x 104 cfu/ml for the first and second sampling respectively and for the total coliform count, 0.18 x 104 cfu/ml and 0.17 x 104 cfu/ml for the first and second sampling respectively. Statistical analysis using student’s T-test was carried out. The mean test values for total heterotrophic bacteria in the first and second samplings were 2.2750E2 and 57.4000 in NDDC hostel and 1.5060E2 and 66.8000 in post graduate hostel. The mean values for total coliform count for the first and second samplings were 1.6220E2 and 24.5000 in NDDC hostel and 99.8000 and 26.9000 in Post graduate hostel. The test revealed that there was no significant difference in the bacterial load of NDDC hostel and Post graduate hostel from the two samplings. In the antimicrobial susceptibility test carried out, results showed that Ciproflox, Reflacine and Tarrivid were most effective against the isolates, whileth the isolates mostly showed resistance against Amplicox, Amoxil and Azithromycin. The bacteria species isolated were Staphylococcus aureus, Escherichia coli, Staphylococcus spp, Bacillus cereus, Klebsiella spp, Bacillus spp. The study demonstrated that significant numbers of E.coli, and staphylococcus as well as other microbes occur in bath towels.

ABSTRACT Termites of the Nasutitermes genus are considered one of the main urban and agroforestry pests in Brazil, where the main method of control is the application of pesticides. The uso of entomopathogenic fungi to reduce the population of this plague in the environment could be use as an alternative. The goal of this study was to evaluate the virulence of isolates native Amazonian fungi belonging to the Tolypocladium endophyticum, Metarhizium anisopliae and Metarhizium marquandii species for the control of Nasutitermes sp. The strains of T. endophyticum (4.439), M. anisopliae (4.443) and M. marquandii (4.472) with their respective isolation codes, were evaluated using suspensions at concentrations of 105, 106, 107 and 108 conidia/mL against the termites. The fungi were characterized to species level by molecular analysis. The greatest virulence was registered with T. endophyticum (4.439), with a mortality of 100 % on the 4th day of treatment for all analyzed concentrations. The M. anisopliae strain (4.443) proved to be efficient, causing a mortality of 100 % on the 7th and 6th days at dilutions of 107 and 108 conidia/mL, respectively. Rates lower than 100 % were registered with M. marquandii (4.472). Therefore, the three fungal strains showed virulence against the termites Nasutitermes sp. In this study, the fungi Tolypocladium endophyticum and Metarhizium marquandii are reported for the first time for the biological control of pests, indicating the potential of native Amazonian fungi for the biological control of thermites Nasutitermes sp.


RESUMEN Las termitas del género Nasutitermes son consideradas una de las principales plagas urbanas y agroforestales de Brasil, donde el principal método de control es la aplicación de pesticidas. Los hongos entomopatógenos pueden ser usados como alternativa para reducir la población de esta plaga en el medio ambiente. Por tanto, el objetivo de este estudio fue evaluar la virulencia de aislados de hongos nativos de suelo amazónico pertenecientes a las especies Tolypocladium endophyticum, Metarhizium anisopliae y Metarhizium marquandii para el control de Nasutitermes sp. Las cepas de T. endophyticum (4,439), M. anisopliae (4,443) y M. marquandii (4,472) con sus respectivos códigos de aislamiento, fueron evaluadas utilizando suspensiones a concentraciones de 105, 106, 107 y 108 conidios/mL contra las termitas. Los hongos se caracterizaron mediante análisis moleculares para confirmar la especie. La mayor virulencia se registró con el hongo T. endophyticum (4,439), con una mortalidad del 100 % al cuarto día de tratamiento para todas las concentraciones analizadas. La cepa M. anisopliae (4,443) demostró ser eficiente, causando una mortalidad del 100 % al sexto y septimo días a las diluciones de 107 y 108 conidios / mL, respectivamente. Se registraron tasas inferiores al 100 % con M. marquandii (4,472). Por tanto, las tres cepas de hongos mostraron virulencia contra las termitas Nasutitermes sp. En este estudio, los hongos Tolypocladium endophyticum y Metarhizium marquandii son reportados por primera vez para el control biológico de plagas, indicando el potencial de hongos nativos de suelo amazónico para el control biológico de termitas Nasutitermes sp.

Objective To compare the differences in pathogenicity and gene expression profiles between adult Schistosoma japonicum isolated from hilly and marshland and lake regions of Anhui Province, so as to provide the scientific evidence for formulating the precise schistosomiasis control strategy in different endemic foci. Methods C57BL/6 mice were infected with cercariae of S. japonicum isolates from Shitai County (hilly regions) and Susong County (marshland and lake regions) of Anhui Province in 2021, and all mice were sacrificed 44 days post-infection and dissected. The worm burdens, number of S. japonicum eggs deposited in the liver, and the area of egg granulomas in the liver were measured to compare the difference in the pathogenicity between the two isolates. In addition, female and male adult S. japonicum worms were collected and subjected to transcriptome sequencing, and the gene expression profiles were compared between Shitai and Susong isolates of S. japonicum. The differentially expressed genes (DEGs) were subjected to Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses. Results The total worm burdens [(14.50 ± 3.96) worms/mouse vs. (16.10 ± 3.78) worms/mouse; t = 0.877, P = 0.392], number of female and male paired worms [(4.50 ± 0.67) worms/mouse vs. (5.10 ± 1.45) worms/mouse; t = 1.129, P = 0.280], number of unpaired male worms [(5.50 ± 4.01) worms/mouse vs. (5.60 ± 1.69) worms/mouse; t = 0.069, P = 0.946], number of eggs deposited in per gram liver [(12 116.70 ± 6 508.83) eggs vs. (16 696.70 ± 4 571.56) eggs; t = 1.821, P = 0.085], and area of a single egg granuloma in the liver [(74 359.40 ± 11 766.34) µm2 vs. (74 836.90 ± 13 086.12) µm2; t = 0.081, P = 0.936] were comparable between Shitai and Susong isolates of S. japonicum. Transcriptome sequencing identified 584 DEGs between adult female worms and 1 598 DEGs between adult male worms of Shitai and Susong isolates of S. japonicum. GO enrichment analysis showed that the DEGs between female adults were predominantly enriched in biological processes of stimulus response, cytotoxicity, multiple cell biological processes, metabolic processes, cellular processes and signaling pathways, cellular components of cell, organelles and cell membranes and molecular functions of binding and catalytic ability, and KEGG enrichment analysis showed that these DEGs were significantly enriched in pathways of vascular endothelial growth factor signaling, glutathione metabolism, arginine and proline metabolism. In addition, the DEGs between male adults were predominantly enriched in biological processes of signaling transduction, multiple cell biological processes, regulation of biological processes, metabolic processes, development processes and stimulus responses, cellular components of extracellular matrix and cell junction and molecular functions of binding and catalytic ability, and these DEGs were significantly enriched in pathways of Wnt signaling, Ras signaling, natural killer cells-mediated cytotoxicity, extracellular matrix-receptor interactions and arginine biosynthesis. Conclusions There is no significant difference in the pathogenicity between S. japonicum isolates from hilly and marshland and lake regions of Anhui Province; however, the gene expression profiles vary significantly between S. japonicum isolates.

Lettuce bacterial leaf spot caused by Xanthomonas campestris pv. vitians is an aggressive disease that is difficult to control. So far there are no reports of the reaction of biofortified lettuce genotypes to different isolates of the bacteria. Thus, the objective was to evaluate the aggressiveness of X. campestris pv. vitians, as well as the reaction of biofortified lettuce genotypes to bacterial spot. Two experiments were performed in two distinct seasons (winter and summer), in greenhouse at the Vegetable Experimental Station of the Federal University of Uberlândia (UFU). The experimental design in both experiments was a randomized block design, in a factor scheme of 5 × 4 (five genotypes and four strains), with four repetitions. Were evaluated the severity and the area under the disease progress curve. In general, the biofortified lettuce 'Uberlândia 10000' was more resistant to most bacterial strains in the summer cultivation, and in the winter period UFU 'Crespa 206'. The commercial cultivar Robusta was the most susceptible to the strains during both seasons. The UFU E125 strain was the most aggressive for most genotypes in both seasons.

Entomopathogenic fungi (EPF) now a possible safer microbial control measure that could be considered as a substitute for chemical control of insect pests. Three EPF viz., Metarihizium anisopliae, Isaria furnosoroseus and Beauveria bassiana were evaluated for their virulence against the grubs of Khapra beetle, Trogoderma granarium (Everts) under laboratory conditions. The isolates were applied by two methods viz., diet incorporation and an immersion method with 3rd instar 20 grubs of T. granarium for each. The virulence of EPF was determined using percent mortality. Significantly higher mortality was observed in M. anisopliae applied through immersion (98.33%) and diet incorporation (93.33%) methods followed by B. bassiana (90.83 and 85.83%, respectively). The mortality caused by I. furnosoroseus was statistically lower in immersion and diet incorporation methods i.e. 81.67 and 73.33%, respectively. Based on the immersion method, all EPF were studied for multiple conidial concentration i.e., 1×104 , 1×105, 1×106, 1×107 and 1×108 under the same in-vitro conditions. All the isolates were pathogenic to grub of T. granarium at the highest conidial concentration. M. anisopliae was proved the most effective virulent resulting in 98.33% mortality of the pest with LT50 4.61 days at 1 × 108 conidial concentration followed by 90.83 and 81.67 percent mortality with 5.07 and 8.01 days LT50, in the application of B. bassiana and I. furnosoroseus, respectively. M. anisopliae showed higher efficacy and could be considered as promising EPF for the development of myco-insecticides against effective biocontrol of T. granarium.


Os fungos entomopatogênicos (FPE) são agora a possível medida de controle microbiano mais segura, que pode ser considerada um substituto para o controle químico de pragas de insetos. Três EPF viz., Metarihizium anisopliae, Isaria furnosoroseus e Beauveria bassiana foram avaliados quanto à sua virulência contra as larvas do besouro Khapra, Trogoderma granarium (Everts) em condições de laboratório. ABSTRACT

This study was designed to identify the pathogen causing soft rot of Pinellia ternata in Qianjiang of Hubei province and screen out the effective bactericides, so as to provide a theoretical basis for the control of soft rot of P. ternata. In this study, the pathogen was identified based on molecular biology and physiological biochemistry, followed by the detection of pathogenicity and pathogenicity spectrum via plant tissue inoculation in vitro and the indoor toxicity determination using the inhibition zone method to screen out bactericide with good antibacterial effects. The control effect of the bactericide against P. ternata soft rot was verified by the leave and tuber inoculation in vitro. The phylogenetic tree was constructed based on the 16 S rDNA, dnaX gene, and recA gene sequences, respectively, and the result showed that the pathogen belonged to the same branch as the type strain Dickeya fangzhongdai JS5. The physiological and biochemical tests showed that the pathogen was identical to D. fangzhongdai, which proved that the pathogen was D. fangzhongdai. The pathogenicity test indicated that the pathogen could obviously infect leaves at 24 h and tubers in 3 d. As revealed by the indoor toxicity test, 0.3% tetramycin, 5% allicin, and 80% ethylicin had good antibacterial activities, with EC_(50) values all less than 50 mg·L~(-1). Tests in tissues in vitro showed that 5% allicin exhibited the best control effect, followed by 0.3% tetramycin and 10% zhongshengmycin oligosaccharide, and their preventive effects were better than curative effects. Therefore, 5% allicin can be used as the preferred agent for the control of P. ternata soft rot, and 0.3% tetramycin and 10% zhongshengmycin oligosaccharide as the alternatives. This study has provided a certain theoretical basis for the control of P. ternata soft rot.

ABSTRACT

Moraxella catarrhalis ( Mca) is a kind of gram-negative diplococcus which can exist in the respiratory tract of the human. It could be a non-symptom diplococcus on the health people. Otitis media occurs when the Mca reaches the middle ear along the eustachian tube. Sometimes the patients could suffer from the acute exacerbation of chronic obstructive pulmonary disease or pneumonia due to lung lesions caused by Mca. Little is known about the pathogenesis of the Mca, which leads to an incomplete understanding of its pathogenicity. This review aims to clarify the relationships between the Mca and the related diseases and the mechanism of the significant virulence factors. We hope to raise awareness of Mca and also provide some ideas for clinical diagnosis of relevant diseases it caused.

Abstract The mortality of the Alabama argillacea and Chrysodeixis includens (Lepidoptera: Noctuidae) larvae caused by the kaolin inert powder and the entomopathogenic fungus Beauveria bassiana were determined under laboratory conditions. Using the caterpillar submersion method, the CG 138 B. bassiana isolate was more pathogenic to A. argillacea than the CG 70, GC 82, ESALQ 634, and ESALQ 645. All five tested isolates caused similar mortality of C. includens. The mortality of first-instar larvae of A. argillacea and C. includens by feeding on leaf-disc impregnated with B. bassiana (CG 138) and kaolin was also determined. Higher A. argillacea mortalities were observed in the B. bassiana (CG 138) treatments, regardless of the presence of kaolin. However, the activity of kaolin + B. bassiana (CG 138) against C. includens was higher than each ingredient alone, indicating an additive action against C. includes larvae. The mortality of A. argillacea and C. includens larvae treated with kaolin + B. bassiana (CG 138) was similar, and the A. argillacea mortality was higher than that of C. includens with kaolin and B. bassiana (GC 138) separated. The treatment kaolin + B. bassiana (CG 138) is promising for the simultaneous management of these two defoliator pests, mainly A. includes. In addition, the monophagous A. argillacea is more susceptible to both kaolin and B. bassiana (GC 138) than the polyphagous C. includens, suggesting that the nutritional ecology plays an important role in the susceptibility of these defoliator species to alternative insecticides.


Resumo A mortalidade de larvas de Alabama argillacea e Chrysodeixis includens (Lepidoptera: Noctuidae), causada pelo pó inerte do caulim e pelo fungo entomopatogênico Beauveria bassiana, foi determinada em condições de laboratório. Usando o método de submersão da lagarta, o isolado CG 138 de B. bassiana foi mais patogênico para A. argillacea que os CG 70, GC 82, ESALQ 634 e ESALQ 645 desse fungo. Os cinco isolados testados causaram mortalidade semelhante de C. includens. A mortalidade de larvas de primeiro ínstar de A. argillacea e C. includens pelo método de alimentação em discos foliares impregnados com B. bassiana (CG 138) e caulim foi, também, determinada. A mortalidade de A. argillacea foi maior nos tratamentos com B. bassiana (CG 138), independentemente da presença do caulim. A atividade do caulim + B. bassiana (CG 138) contra C. includens foi maior que cada ingrediente isolado, indicando ação aditiva contra larvas desse Lepidoptera. A mortalidade de larvas de A. argillacea e C. includens, com caulim + B. bassiana (CG 138), foi semelhante e as de A. argillacea maiores que as de C. includens com caulim e B. bassiana (GC 138) isoladamente. O tratamento caulin + B. bassiana (CG 138) é promissor para o manejo simultâneo dessas duas pragas desfolhadoras, principalmente A. includens. Além disso, a monófaga, A. argilacea, é mais suscetível ao caulim e à B. bassiana (GC 138) que a polífaga, C. includens, sugerindo que a ecologia nutricional desempenha papel importante na suscetibilidade dessas espécies desfolhadoras a inseticidas alternativos.

ABSTRACT

Abstract: human immunodeficiency virus type 1 (HIV-1) is the etiological agent of acquired immunodeficiency syndrome (AIDS), a pandemic with high economic and social costs. The envelope glycoprotein (ENV) of the virus mediates the infectious process by binding to and entering the host cell, one of the main target components of studies since its discovery. Its endodomain or C-terminal tail (CTT) participates in late replicative cycle processes, such as intracellular trafficking, activation, and cell death, which occurs because it interacts with multiple cellular factors through motifs or signal sequences present throughout its structure. Although these interactions have not been fully understood at specific levels, studies over more than three decades leave no doubtthatthis domain plays a fundamental role in the biology of the virus and probably the development of the disease. This review describes the studies carried out to date that demonstrate the importance of the CTT, focusing on the motifs responsible for its interactions and its possible roles in the pathogenicity of the infection.


Resumen: el virus de la inmunodeficiencia humana tipo 1 (VIH-1) es el agente etiológico del síndrome de inmunodeficiencia adguirida (SIDA), una pandemia con altos costos económicos y sociales. La glicoproteína de la envoltura (ENV) del virus media el proceso infeccioso al unirse a la célula huésped y entrar en ella, uno de los principales componentes objetivo de los estudios desde su descubrimiento. Su endodominio o cola C-terminal (CTT) participa en procesos tardíos del ciclo replicativo, como tráfico intracelular, activación y muerte celular, lo que ocurre porque interactúa con múltiples factores celulares a través de motivos o secuencias señal presentes en toda su estructura. Aunque estas interacciones no se han entendido completamente a niveles específicos, los estudios durante más de tres décadas no dejan dudas de que este campo juega un papel fundamental en la biología del virus y probablemente en el desarrollo de la enfermedad. Esta revisión describe los estudios realizados hasta la fecha que demuestran la importancia de la CTT, centrándose en los motivos responsables de sus interacciones y sus posibles roles en la patogenicidad de la infección.

A glicoproteína do envelope (ENV) do vírus media o processo infeccioso ligando-se e entrando na célula hospedeira, um dos principais componentes alvo dos estudos desde sua descoberta. Seu endo domínio ou cauda C-terminal (CTT) participa de processos do ciclo replicativo tardio, como tráfego intracelular, ativação e morte celular, que ocorre porque interage com múltiplos fatores celulares por meio de motivos ou sequências-sinal presentes em toda a sua estrutura. Embora essas interações não tenham sido totalmente compreendidas em níveis específicos, estudos ao longo de mais de três décadas não deixam dúvidas de que esse domínio desempenha um papel fundamental na biologia do vírus e provavelmente no desenvolvimento da doença. Esta revisão descreve os estudos realizados até o momento que demonstram a importância da CTT, com foco nos motivos responsáveis por suas interações e seus possíveis papéis na patogenicidade da infecção.

RESUMEN La enfermedad por coronavirus 2019 (COVID-19) es causada por un nuevo betacoronavirus conocido como síndrome respiratorio agudo severo coronavirus-2 (SARS-CoV-2). Para el 22 de junio del 2021, el número de casos confirmados en todo el mundo había superado los 178 millones, con más de 3 millones de muertes. La fisiopatología de la COVID-19 a partir de la infección por SARS-CoV-2 no está del todo dilucidada. En el presente artículo se exponen los hallazgos encontrados después de la búsqueda en la literatura científica realizada en la base de datos PubMed entre octubre de 2020 y abril de 2021 en la cual se incluyeron 71 artículos, con el objetivo de la revisión fisiopatológica completa, detallada y actualizada del SARS-CoV-2, abordando temas como la caracterización y ciclo de vida del virus, el mecanismo de transmisión, la cinética viral y la respuesta inmune, junto con la dinámica fisiopatológica de la infección. ABSTRACT Coronavirus disease 2019 (COVID-19) is caused by a new betacoronavirus named as Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2). On June 22nd, 2021, the number of confirmed cases worldwide exceeded 178 million, resulting in more than 3 million deaths. The pathophysiology of COVID-19 from the infection of SARS-CoV-2 is not entirely elucidated. This review presents the findings after the research in the scientific literature carried out in the PubMed database between October 2020 and April 2021, in which 71 articles were included, with the aim of a complete, detailed and updated pathophysiological review of SARS-CoV-2, addressing issues such as the characterization and life cycle of the virus, the transmission mechanism, viral kinetics and immune response, along with the pathophysiological dynamics of the infection.
